Manganese Superoxide Dismutase (MnSOD) is a crucial human mitochondrial antioxidant enzyme that prevents damage to cellular structures such as mitochondrial DNA and proteins by catalyzing the dismutation reaction of the superoxide radical (O2•−) into oxygen (O2) and hydrogen peroxide (H2O2). In situations of oxidative stress, O2•− reacts with nitric oxide (•NO), generating an intermediate product called peroxynitrite (ONOO-), which can inactivate MnSOD, resulting in the loss of its function and contributing to the pathogenesis of multiple diseases. The COPASI software was used to simulate chemical reactions and determine the formation of peroxynitrite and its link to MnSOD inactivation. Peroxynitrite formation was confirmed even under conditions where it was previously believed unlikely due to the high enzymatic catalysis rate of MnSOD. Its formation leads unequivocally to the enzyme's complete inactivation through a negative feedback mechanism.
